An Electron Microscopic Study of Liver Cell Following Common Bile Duct Ligation in Rat


Abstract
To study changes of fine structure and some aspects of function of the liver following bile duct ligation, electromicroscopic as well as light microscopic examination for morphologic study and SGPT, total bilirubin and alkaline phosphatase as liver function tests were done in 12 hours, 1 day, 3 days and 30 days after commen bile duct ligation in rats. And findings obtained could be summarised as follows.
1. Serum trnasmainase (SGPT), alkaline phosphatase and total bilirubin levels rose acutely and became peak level in 1-3 days following common bile durt ligation and thereafter declined progressively becoming almost normal in 30 days.
2. In light microscopic examination of the liver, there noted scattering focal necrosis with hemorrhage already in 12 hours which became rarer in 10 and 30 day groups while granulation and septal fibrodis became increasingly prominent. (:ytological examination showed groups of degenerating liver cells with inflammatory cell collectio~~ in 12 hours group which was compatible with light microscopic findings. Anisokaryosis observed, liy cytology was more apparent than by histology.
3. In electromicroscopic examination, there noted ER changes including dilatation, fragmentation and increased free ribosome, and changes in Golgi bodies and mitochondria up to 10 days after common bie duct ligation, but showed nearly normal pattern in 30 days group.
Bile canaliculi showed only minimal changes.. There were no evidence of bile stasis in liver cells and evidence of bile regurgition through shunts between bile canaliculi and sinusoids.
4. Remarkable elevation of serum bilirubin level i~ispite of minimal or negligible morphological evidence of bile stasis in bile canalicular system may suggest the possible explanation of h.yperbilirubinemia by liver cell dysfunction with disti.trbed microsomal biotransformation system which began and became serious before the morpholgical changes in biliary system become evident.
